Nuclear Envelope Integrity in Health and Disease: Consequences on Genome Instability and Inflammation

چکیده
The dynamic nature of the nuclear envelope (NE) is often underestimated. NE protects, regulates, and organizes eukaryote genome adapts to epigenetic changes its environment. morphology characterized by a wide range diversity abnormality such as invagination blebbing, it diagnostic factor for pathologies cancer. Recently, micronuclei, small nucleus that contains full chromosome or fragment thereof, has gained much attention. micronuclei prone collapse, leading DNA release into cytoplasm with consequences ranging from activation cGAS/STING pathway, an innate immune response, creation chromosomal instability. discovery those mechanisms revolutionized understanding some inflammation-related diseases origin complex rearrangements, observed during initiation tumorigenesis. Herein, we will highlight complexity biology discuss clinical symptoms in NE-related diseases. interplay between immunity, genomic instability, leakage could be major focus future years explain lead new classes therapeutics.

15 صفحه اولNuclear envelope rupture drives genome instability in cancer
The nuclear envelope, composed of two lipid bilayers and numerous accessory proteins, has evolved to house the genetic material of all eukaryotic cells. In so doing, the nuclear envelope provides a physical barrier between chromosomes and the cytoplasm. Once believed to be highly stable, recent studies demonstrate that the nuclear envelope is prone to rupture. These rupture events expose chromo...

15 صفحه اولNuclear envelope influences on genome organization.
The nuclear periphery is a specialized environment in the nucleus that contributes to genome organization and correspondingly to gene regulation. Mammalian chromosomes and certain genes occupy defined positions within the nucleus that are heritable and tissue specific. Genes located at the nuclear periphery tend to be inactive and this negative regulation can be reversed when they are released ...
